摘要: 将一种肠毒素抗体共价耦联于自组装的金电极表面 ,利用原子力显微镜识别抗原抗体在多种实验条件下的形貌并通过统计数据进行定量分析 .用作控制实验的是非特异性牛血清白蛋白(BSA)抗原以及低浓度的SEC1抗原 .另外 ,酸性溶液可以打破抗原与抗体之间的结合从而使得生物电极可以被重复使用 .电化学实验支持了AFM的测试结果

Abstract: The goals of this work were to demonstrate the ability of atomic force microscopy (AFM) to detect various immunological processes between antibodies and antigens.A sensor was designed by covalently linking the antibody of staphylococcal enterotoxin Cl (SEC1) on the self-assembled gold surface. This sensor permitted detection of antibody-antigen recognition events which were highly correlated with the topography under different conditions. The statistic data of AFM were analysed. As control experiments, nonspecific antigens and lower concentration SEC1 antigens were directly observed. In addition, an acidic solution was used to break the antigen-antibody pair and made the sensor reuse. Electrochemical evidence was provided to support the results obtained with AFM.
